Expression profile of genes involved in osteoclast differentiation derived from bone marrow



Abstract
The cDNA array technique was employed to analyse gene expression patterns during osteoclast differentiation from mouse bone marrow precursor cells. The following conclusions were drawn from the obtained results.
1. Cytokine receptors such as interleukin-7 receptor alpha and insulin-like growth factor 1A, hormone precursors such as proopiomelanocortin, signaling molecules such as interferon regulatory factor 1 and PI3-K p85 alpha, transcription factors such as c-fos proto-oncogene, and cell cycle regulatory genes such as TIS21 early response gene increased during osteoclast differentiation.
2. TCRA enhancer-binding factor-interacting protein 1, proliferation-associated protein 1, prothymosin alpha, small inducible cytokine subfamily A member 7, prothymosin beta 4, and rho GDP dissociation inhibitor beta decreased during osteoclast differentiation. These genes have been show to either mediate signaling pathways or regulate transcription. However, their involvement in osteoclast differentiation has not been reported. Some genes involved in cell cycle regulation such as G2/M-specific cyclin G, G2/M-specific cyclin A2. G2/M-specific cyclin B2, and cyclin-dependent kinase inhibitor 2D also decreased during osteoclastogenesis.
